as for layers, I sometimes do that, but only for punctual waves or really weak points. Otherwise I just do a single proper layer

I've found that having soldiers in stone towers allows for a very chunky defense, so if you start using advanced units for horde clearing if you build a bunch of stone towers and fill with soldiers you get effectively 4-5 layers of wall, additionally if the defenses get breached soldiers are vastly better when moving from a safe area of the wall to reinforce the breach
I've found Titans to be extremely powerful and easily be worth their cost-if you micro them so that you are getting full value out of their AOE instead of the auto-target's about half of their damage output you can start deleting hordes rapidly-faster than with Thanatoses. while being properly micro'd a couple of Titans can easily hold a smaller chokepoint against an infinite amount of normal-ish zombies. I've found having 1-3 groups of two to send to areas of the defense where zombies are clumping up to remove clusters of zombies to be the best use out of them
